Colourimetric method for the determination of β-Glucanase
in malt, foodstuffs and fermentation products

Principle:
(β-glucanase)
(1) Azo-Barley β-glucan (polymer) → Azo-barley β-glucan
(fragments)

(2) Add alcohol; centrifuge to remove polymeric Azo-barley
β-glucan

(3) Measure the absorbance of the supernatant solution

Reaction time: ~ 30 min
Detection limit: 100 U/kg of malt

     β-葡聚糖酶作用方式不同，可分为内切型和外切型。前者存在于谷物种子、某些真菌和某些细菌中，能催化水解谷物细胞壁中的β-葡聚糖，其中包括内切型β-1,4-葡聚糖酶、内切型β-1,3-葡聚糖酶。后者存在于谷物种子中，其中又包括外切型β-1,4-葡聚糖酶、外切型β-1,3-葡聚糖酶。
在工业上采用细菌（如枯草芽孢杆菌）、曲霉（如黑曲霉）、青霉（如伊氏青霉）生产。在啤酒生产原料大麦及其麦芽中，含本品较多。在啤酒生产中采用β-葡聚糖酶，可提高麦汁的过滤速度和得率，从而保证啤酒质量。也可用于饴糖、麦芽糖浆生产和用作饲料添加剂。

原理：

(citrate lyase)


(1) Citrate → oxaloacetate + acetate


(L-malate dehydrogenase)


(2) Oxaloacetate + NADH + H+ → L-malate + NAD+


(D-lactate dehydrogenase)
