    甲苯胺蓝，CAS号：92-31-9，又名碱性蓝 17，英文名Toluidine blue。 甲苯胺蓝是常用的人工合成染料的一种，属于醌亚胺染料类，含有两个发色团，还含有两个助色团，为碱性染料。甲苯胺蓝中的阳离子有染色作用，组织细胞的酸性物质与其中的阳离子相结合而被染色。可染细胞核使之呈蓝 色；肥大细胞胞质内含有肝素和组织胺等异色性物质遇到甲苯胺蓝可呈异染性紫红色，可用于尖锐湿疣的初筛及肥大细胞的检测，例如色素性寻麻疹。

    使用方法

    染色方法:
    1.甲苯胺蓝液: 甲苯胺蓝0.5g蒸馏水定容至100ml
    2.冰醋酸液: 冰醋酸0.5ml蒸馏水定容至100ml
    染色步骤
    (一)
    1.组织切片常规脱蜡至水
    2.入甲苯胺蓝液30min
    3.稍水洗
    4.入冰醋酸液分化，直到胞核和颗粒清晰。
    5.稍水洗，用冷风吹干
    6.二甲苯透明，中性树胶封固
    (二)
    ①切片脱腊至水。
    ②蒸馏水浸洗3次。
    ③0.1%甲苯胺蓝液浸染10min。
    ④水洗，洗去多余染液。
    ⑤各级酒精脱水。
    ⑥二甲苯透明，中性树胶封固。
    试剂的配制
    称取0.1g甲苯胺蓝，溶于100ml 0.1M醋酸盐缓冲液中，置室温5天后，即可使用染液于室温下可保存3个月，若放于4℃冰箱则可保存半年左右，但该液的染色效果在早期为最佳，若染液存放时间较长，则染色偏淡，应适应延长染色时间。
