Avian Leukosis Virus is a common naturally occurring neoplastic disease affecting chickens. AL is of great economic significance not only because of tumor production, but decreased egg production and increased mortality. Due to its long incubation period, AL affects mostly breeders and commercial egg layers. Enlargement of the bursa and liver may be detected by palpitation in advanced cases.1 Avian viruses in serum can be detected by the Enzyme Linked Immunosorbant Assay (ELISA). The levels of viral protein p27 in serum and egg albumen, as well as the disease status of entire flocks can be monitored using ELISA.

Any p27 viral proteins present in the serum will bind to the -p27 on the microwell forming a p27 antibody-antigen complex. After washing away any unbound material, anti-p27 IgG Alkaline Phosphatase conjugate is added to the microwell. The conjugate will bind to the p27 antibody-antigen complex. After incubation any unbound conjugate is removed by washing. Enzyme substrate is then added to the microwell. The substrate reacts with any Alkaline Phosphatase present and forms a yellow product. In the final step the reaction is stopped, fixing the color. The intensity of the color is measured photometrically at 405 – 410 nm.

Newcastle Disease (ND) has worldwide distribution and Newcastle Disease Virus is by far one of the most important pathogens of all birds. The symptoms of ND include respiratory distress, diarrhea, cessation of egg production, depression, edema of the head, face, and wattles, nervous signs, and death. Birds may display all, some, or none of these symptoms, Newcastle Disease can vary from bird to bird and cause sudden death with 100% mortality to subclinical disease.1 Antibodies to avian viruses in serum can be detected by the Enzyme Linked Immunosorbant Assay (ELISA). This test allows the quantitative flow cytometric determination of the chemotactic function of neutrophilic granulocytes. In analogy to the Boyden chamber technique, this kit contains multiwell plates and precoated cell culture inserts for chemotaxis studies, the chemotactic peptide N-formyl-Met-Leu-Phe (fMLP), an antibody reagent, counting beads and necessary reagents. It allows determination of the number of neutrophils which have migrated through cell culture inserts towards a concentration gradient of the chemoattractant fMLP.
Also, the decrease of expresssion of the cell adhesion molecule LECAM-1 can be analysed together with the shape-change of the cells by analysing the changes of the forward scatter signals.
