The Acetic Acid GK format test kit is for use with auto-analysers and is suitable for the specific measurement and analysis of acetic acid (acetate) especially in wines, fruit juices, beverages and food products.
As part of Megazyme’s overall commitment to providing the highest quality products, we have developed this acetic acid kit that provides a specific and rapid assay for use with auto-analysers. The kit assay is based on the conversion of NAD+ to NADH and therefore provides a positive reaction (increase in absorbance) which offers a more robust assay.
The reagents, as supplied, are stable for a minimum of 2 years and the prepared reagents are stable for a minimum of 1 week (on-board stability). In addition, the prepared reagents can be stored frozen for longer term stability (see K-ACETGK Booklet for more details).

The Acetic Acid (Acetate Kinase Manual Format) test kit is suitable for the measurement and analysis of acetic acid in food and beverages.
This rapid and reliable manual acetic acid kit is simple to perform (only two absorbance readings required), and because a true end-point is measured, does not involve complicated calculations like other kits. This product is very stable both during storage and use (> 2 years), has extended linearity (compared to ACS based kits), contains PVP to prevent tannin inhibition, and is performed at a relatively low pH (7.4), thus minimising ester hydrolysis related interference. This method is suitable for the measurement of acetic acid/acetate in foods, beverages and other materials.

The Available Carbohydrates Assay Kit method is suitable for the determination of available carbohydrates (AVCHO) comprising *total digestible starch (TDS) plus maltodextrins, sucrose, D-glucose, D-fructose and lactose. New Improved method receiving ‘First Action’ status: AOAC 2020.07. This method is designed to simulate in vivo conditions in the human small intestine (i.e. a 4 h incubation time with PAA + AMG) in parallel with recent advances in Dietary Fiber (DF) methodology (K-RINTDF: AOAC Method 2017.16) and in accordance with the new (physiological based) definition of DF announced by Codex Alimentarius in 2009. Also, sucrose is hydrolysed with a specific “sucrase” enzyme which (unlike invertase which has been used traditionally for this reaction) has no action on fructo-oligosaccharides (FOS).
* Total digestible starch (TDS) is defined as starch that is digested in a 4 h period and is part of the carbohydrate that is available for digestion and absorption in the human small intestine.

Cellulase Activity Assay Kit.
The CellG5 assay reagent for the measurement of endo-cellulase (endo-1,4-β-glucanase) contains two components; 1) 4,6-O-(3-Ketobutylidene)-4-nitrophenyl-β-D-cellopentaoside (BPNPG5) and 2) thermostable β-glucosidase. The ketone blocking group prevents any hydrolytic action by the β-glucosidase on BPNPG5. Incubation with an endo-cellulase generates a non-blocked colourimetric oligosaccharide that is rapidly hydrolysed by the ancillary β-glucosidase. The rate of formation of 4-nitrophenol is therefore directly related to the hydrolysis of BPNPG5 by the endo-cellulase. The reaction is terminated and the phenolate colour is developed on addition of Tris buffer solution (pH 9.0).
The CellG5 assay represents a huge step forward in the methodology for the measurement of cellulase that traditionally relied on substrates such as CM-cellulose, Avicel, cellooligosaccharides, filter paper or dyed polysaccharides including CMC Congo red or cellulose azure.

The CellG3 assay reagent for the measurement of endo-cellulase (endo-1,4-β-glucanase) contains two components;
1) 4,6-O-benzylidene-2-chloro-4-nitrophenyl-β-D-cellotrioside (BCNPG3) and 2) thermostable β-glucosidase. The benzylidene blocking group prevents any hydrolytic action by the β-glucosidase on BCNPG3. Incubation with an endo-cellulase generates a non-blocked colourimetric oligosaccharide that is rapidly hydrolysed by the ancillary β-glucosidase. The rate of formation of 2-chloro-4-nitrophenol is therefore directly related to the hydrolysis of BCNPG3 by the endo-cellulase. The reaction is terminated and the phenolate colour is developed on addition of Tris buffer solution (pH 9.0).
Please note that a new assay kit (K-CellG5) is now available for the measurement of endo-cellulase. The CellG5 reagent contains a cellopentaose core and exhibits vastly improved sensitivity for some cellulases. In addition, the exchange of the benzylidene blocking group in CellG3 for 3-keto-butylidene in CellG5 improves the substrate’s water solubility significantly, allowing for a reduction in the concentration of DMSO required in the assay. As DMSO is known to inhibit certain cellulases, this is another benefit in using CellG5. Megazyme now recommends the use of K-CellG5 for all assays for the measurement of endo-cellulase.
